Harnessing Natural Killer Cell-Mediated Innate Immune Responses for Cancer Treatment: Advances and Challenges

Natural killer (NK) cells are a gatekeeper of the body’s innate defense system against cancers and infections. A growing body literature from us others finds that NK promote anti-cancer immune surveillance, defects in cell development associated with poor clinical prognosis cancers. In preclinical studies, were found to drive tumor regression delay relapse. Because potentially less damaging easier develop than T cell-based therapies, efforts being made improve cytotoxicity vivo persistence for use as an adoptive, off-the-shelf immunotherapy. this review, we discuss how tumor-intrinsic -extrinsic factors suppress cancer microenvironment. We also outline current strategies restore surveillance challenges facing therapies.
